This study evaluates the analgesic and anti-inflammatory effects of Ganoderma lucidum ethanol extract using acetic acid-induced writhing and formalin-induced paw-licking models in mice. Ganoderma lucidum, a medicinal mushroom with a long history in traditional medicine, was tested at doses of 100, 200, and 400 mg/kg, alongside a control (distilled water) and aspirin (standard) in twenty-five mice. Results showed a dose-dependent reduction in pain, with 400 mg/kg achieving 84.07% inhibition in the writhing test—surpassing aspirin’s 22.12%. In the formalin test, significant anti-inflammatory effects were observed, particularly in the late phase (55.44% inhibition at 400 mg/kg). These findings validate Ganoderma lucidum’s traditional use and its potential as a natural analgesic and anti-inflammatory agent. Further studies should optimize dosage, enhance bioavailability, and explore clinical application.
