EVALUATION OF ANTIASTHMATIC ACTIVITY OF KOLAVIRON EXTRACT AND MUC5AC GENE EXPRESSION IN OVALBUMIN-INDUCED ASTHMA IN GUINEA PIG (CAVIA PORCELLUS)

ABSTRACT

Kolaviron is a complex mixture of bioflavonoid and polyphenols isolated from the seed of Garcinia Kola Heckel commonly used in African Traditional Medicine as a therapy for throat infections, liver disorder and as an aphrodisiac and fertility enhancing substances. A number of studies have confirmed the anti-oxidative and anti-inflammatory effect of kolaviron in chemically induced toxicity, animal models of diseases and in cell cultures. In this study, the anti-asthmatic effect of kolaviron on ovalbumin induced asthma and MUC5AC gene expression in guinea pig animal model was evaluated. Anti-asthmatic activity was analyzed using screening protocol by Bramley et al., (1996) method modified by Salami et al., (2013) while the MUC5AC gene expression was analyzed using polymerase chain reaction (PCR)and agarose gel electrophoresis. In this study, Kolaviron at all dose level significantly caused an increase in the onset of preconvulsive dyspnea on ovalbulmin induced asthma in guinea pigs (*P<0.05, ***P<0.001) when compared to control. Also kolaviron extract in this study, shows that at 25 mg/kg significantly down-regulated MUC5AC gene in ovalbumin induced asthma in guinea pigs when compared negative control (distilled water) (*p < 0.05) and when compared to positive control (albumin) (# p < 0.05) also kolaviron extract (at 50 mg/kg and 100 mg/kg ) and salbutamol (0.5 mg/kg) significantly up-regulated MUC5AC gene in ovalbumin induced asthma in guinea pigs when compared negative control (distilled water) (*p < 0.05) and when compared to positive control (albumin) (# p < 0.05). Conclusively kolaviron extract possess anti-asthmatic activity, thus the use of this extract is recommended for the management of asthma.
