EFFECTS OF UTAZI (GONGRONEMA LATIFOLIUM) EXTRACT ON GATA1 AND STAT5 EXPRESSION GENE

ABSTRACT

Gongronema latifolium a plant traditionally used in various traditional medicines for its health benefits, has attracted attention due to its reported antioxidant, anti-inflammatory, and cytoprotective properties.The aim of this study is to investigate the effects of Utazi extract on GATA1 and STAT5 in mediating haemopoiesis in albino Wistar rats. A total of twenty (20) male adult albino wistar rats were selected into five (4) groups. The groups were control group, 100mg/kg of GroupB, 200mg/kg of Group C, and 300g of Group D. RNA extraction And semi-quantitative polymerase chain reaction (PCR) was used to isolate RNA from rat bone marrow, Complimentary DNA was synthesized and was subsequently amplified using polymerase chain reaction. Gel electrophoresis was used to determine the bands of the genes. The findings revealed that the mRNA expression mRNA expression of STAT-5 was significantly higher in the treated group when compared to control (p<0.05).Groups C showed significantly lower expression of STAT-5 when compared to the untreated group (p<0.05). The mRNA expression of GATA-1 was significantly higher in the treated groups when compared to control (p<0.05). In conclusion, Gongronema latifolium exerted a concentration-based increase and decrease in STAT5 and GATA1 expressions. These results suggest that Gongronema latifolium could influence haematopoeisis by modulating the expression of STAT-5 and GATA-1
