EFFECTS OF HYDROCORTISONE AND MONTELUKAST IN REACTIVE OXYGEN SPECIES: SUPEROXIDE DISMUTASE, CATALASE, MALONDIALDEHYDE, GLUTATHIONE PEROXIDASE AND TOTAL PROTEIN IN ASTHMA-INDUCED WISTER RATS

ABSTRACT

Asthma is a prevalent chronic disease of the respiratory system. Montelukast and hydrocortisone have been reported to discontinue the airways from narrowing (caused by inflammation), however research are few regarding the anti-asthmatic role of Montelukast and hydrocortisone. This study was aimed at investigating the effect of hydrocortisone and montelukast in reactive oxygen species (SOD, CAT, MDA, GPx) and total protein in asthma induced wistar rats. 25 albino Wistar rats weighing between 200-250g were divided into two (2) groups; the Control group and the Test group. All the groups consisted of five (5) rats each (n=5). The control group received normal rat chow and water throughout the experimental period while the test groups were exposed to concentrations of Ovalbumin (OVA, egg albumin grade II) and aluminium hydroxide The Control group was further subdivided into two groups; Control group and negative control group. The test group was further divided into three groups and treated with anti-asthmatic drugs; Montelukast group, Hydrocortisone group and Montelukast/Hydrocortisone group. Asthma was induced by sensitizing the rats with ovalbumin (100mg/kg intraperitoneally) on day 0, 7 and 14 and was challenged with OVA (0.5% w/v absorbed in aluminium hydroxide) three times a week for four weeks. During the period of the challenge, test groups were treated with montelukast (2mg/kg) and hydrocortisone (3mg/kg) and both montelukast and hydrocortisone (2mg/kg and 3mg/kg). On day 28, the rats were sacrificed and the samples taken to laboratory for Analysis. All the data obtained from the experiments are expressed as ± Standard Error of Mean (SEM). Statistical analysis performed by one-way analysis of variance (ANOVA) for accessing differences amongst multiple groups, showing effect of hydrocortisone and montelukast on total protein in asthma induced wistar rats. Result obtained from this study showed no statistically significant difference in Total protein, SOD, CAT, MDA, and GPx among the different groups. In conclusion, the study revealed that hydrocortisone & montelukast had no significant effect on SOD, CAT, MDA, GPx and total Protein.
