EFFECT OF Gongronema latifoluim ON DIBUTYL PHTHALATE-INDUCED CHANGES IN THE ANTIOXIDANT STATUS OF RAT KIDNEY

ABSTRACT

Methanol extract of Gongronema latifolium leaves was investigated for its ameliorative effect on di-butyl phthalate (DBP) induced toxicity in male albino wistar rats of weights 192.4g to 210.4g. Four groups of rats (seven per group) were used in a 14 days study. Group 1 served as the experimental control, Group 2 was given 500mg of di-butyl phthalate orally by gavage for 14 days. Group 3 and group 4 were given 500mg of di-butyl phthalate in addition to oral administration of Gongronema latifolium ecxtract (400mg/kg body weight and 500mg/kg body weight respectively). 500mg/kg body weight of DBP increased the weight of rats to levels that were not significantly different (P>0.05) from control. However, increasing the dose of extract from 400mg/kg body weight to 500mg/kg body weight decreased the weights to levels that were significant (P<0.05). Oxidative stress was estimated by kidney superoxide dismutase, catalase, and malondialdehyde. Administration of methanolic extract of Gongronema latifolium leaves for 14 days significantly (P<0.05) ameliorated the change in the biochemical parameters induced by dibutyl phthalate when compared to the DBP treated groups. However, the activites of catalase (CAT) and superoxide dismutase (SOD) were reduced in levels that were not significant (P>0.05) in the kidney. Increasing the dose of the extract from 400mg/kg body weight to 500mg/kg body weight resulted in significant increase (P<0.05) in the catalase activity, but decrease in MDA concentration. The concentration was increased to levels that were not significantly different from control. This suggested that methanol extract of Gongronema latifolium leaves might have a protective action against Dibutyl phthalate induced renal toxicity due to its bioactive components.
