EFFECT OF ETHANOL EXTRACT OF Anthocleista djalonensis STEM ON THE HAEMATOLOGICAL INDICES AND HISTOLOGY OF THE KIDNEY IN PAROXETINE-INDUCED SEXUAL DYSFUNCTION IN MALE WISTAR RATS

ABSTRACT

This study examined the effect of ethanol extract of Anthocleista djalonensis stem on the haematological indices and histology of the kidney in paroxetine-induced sexual dysfunction in male Wistar rats. This research work was carried out on 35 male Wistar rats which were divided into seven groups; A-G. (n=5). Group A and G served as positive and negative control respectively. The ethanol extract of the stem of Anthocleista djalonensis was evaluated at 100mg/kg and 200mg/kg through oral routes. Group B-G were induced with sexual dysfunction using 200 mg/kg of paroxetine for 7 days. Tadalafil (80mg/kg) was used as a standard reference drug to treat sexually dysfunctional rats. Group A was given only water and feeds, group B and C were treated with 100 mg/kg and 200 mg/kg of ethanol extract of Anthocleista djalonensis respectively, group F was administered with tadalafil and group G was not treated. Treatment lasted for 21 days, after which the rats were sacrificed and blood samples were collected from each rat via cardiac puncture for haematological test and kidney of each rat was harvested for histopathological investigations. The haematological analysis were done using haematological analyzer (sysmex). The parameters determined include: red blood cells (RBC), haematocrit (HCT), mean cell volume (MCV), mean cell hemoglobin ( MCH), mean cell hemoglobin concentration (MCHC), white blood cells (WBC), haemoglobin (HGB), lymphocytes (LYMPH), monocytes ( MON), platelet (PLT) and granulocytes (GRAN). This study's results showed a significant reduction in white blood cell count values by the ethanol extract of Anthocleista djalonensis and tadalafil p (<0.05). RBC, GRAN, HGB, HCT, MCV, MCH and PLT showed no significant difference in all the treated groups when compared to Nthe control groups (p>0.05). Paroxetine caused a significant decrease in MCV and a significant increase in MCHC. P (<0.05). The histopathology examination of the kidney showed abnormal tubules at low dose of the extract. To sum up, the extract may be toxic to the kidney by affecting it functioning. Also, the extract may weaken the body immune system of the rat.
