ABSTRACT

River water contain different kinds of biological pollutants, such as bacteria and fungi. This research was carried out to evaluate the disinfection potential of hydrogen and chlorine on contaminated water. Standard bacteriological methods were used to enumerate the total bacterial and fungal count using pour plate methods after serial dilution. The bacterial isolates were characterized and identified using morphological and biochemical methods and sugar fermentation test. The percentage distribution and frequency of the isolates were evaluated using statistical method. The total heterotrophic bacteria obtained in chlorine treated water was 23.50 x 10 4±1.00 cfu/ml and that of hydrogen peroxide was 22.0 x 104±1.55 cfu/ml, while the none treated water sample was 84.0 x 104±1.33 cfu/ml. The total coliform bacteria was also analyzed based on the treatment. From the result the total coliform for chlorine treatment was 17.0 x 103±2.55 cfu/ml, and hydrogen peroxide was 10 x 103±1.33 cfu/ml while the none treated river water coliform count was 40.0 x 103±2.00 cfu/ml. Same treatment procedure was repeated for fungal analysis where chlorine treatment was (log10 cfu/ml) 20.0 x 104±1.00, hydrogen peroxide treated 18 x 104±1.30 and the none treated sample was 44.0 x 104±3.00 (log10 cfu/ml) There was a significant difference (p<0.05) in the bacterial count for the treated and non-treated water. Based on the morphological and biochemical characteristics the bacteria isolated from the river water in general (treated/untreated) were identified as Bacillus sp, Escherichia coli, Klebsiella sp, Pseudomonas sp, Staphylococcus sp and Salmonella sp while that of fungi include; Aspergillus niger, Penicillium sp and Mucor sp. The antibiotic sensitivity test revealed that all isolates found in this study were susceptible to Gentamicin and Ciprofloxacin but on the other hand were resistance to metronidazole and Colistin.
